Take a look at the (just revised) documentation on Importing and Uploading Assets.  That should help you get started.  If you have a more specific issue, please feel free to post another comment!

I went through that-

But now when I click on "Upload" it says-

"File Does Not Exist- Would you like to create?"

I went through the setups, I don't have ASSET EDITOR, and I can't change permissions even in Admin/Admin

I can't seem to find a way to synchronize folders, and make it so I can access the UPLOAD page, or uploader...

Are you able to get into the Group management area?

 Yes- using the Admin Toolbar in the top Right hand corner- there's a Group Editor.

If you go into the Group Editor and click on the Administrators group, you should see a section of permissions.  Check the "Edit Assets" and "Uploads" checkboxes, and be sure to click the "Save Permissions" link at the bottom of the permissions box.

You should then be able to go back out to your archive (you can get there by selecting it from the Digital Assets menu) and in the Tools menu in the green header bar you should now have access to the Asset Editor tool where you can upload single assets or mount a folder of them. 

I'm guessing that you were trying the Upload link that is in the main header of the page, which obviously doesn't work any more.  We will be sure that the link is removed for our next release.
